An 1869 game between Rutgers and Princeton, played in New Brunswick, N.J., is considered the first intercollegiate football game, so that may be as good a "birthplace" as any -- even though by all accounts, that game resembled soccer much more than it did Rugby or gridiron football.

American football slowly evolved out of rugby in the mid-19th century, after a number of Ivy League colleges playing their own versions of football met in the 1870s to standardize their rules. In the 1880s, Walter Camp came up with the idea of a line of scrimmage to replace the rugby scrum, and he developed a down-and-distance system and the first scoring system.

So there was no single place where it was really invented, although it was mostly played on the East Coast and eventually spread to the Midwest.
