The plug is threaded with an O-ring seal. It also has a fabric seal that allows air to pass through it without letting water in. Since it is a hollow board, pressure from temperature and elevation changes needs to be equalized, and the fabric seal does just that. You should never have to touch it unless either the seal leaks, or you get a crack in the board. In this case you can unscrew it and drain the board.
