When a punter fakes a punt runs and get tackled is that a penalty for the defense?

This is not a penalty. Until the ball has physically left the foot of the punter, he is eligible to be hit. Once it has left, however, any contact may be determined by the ref to be roughing the kicker.
In the case where he runs with it, clearly it has not been kicked so it is legal to tackle him.
