Cornelius McGillicuddy, better known as Connie Mack, holds the MLB record for most career losses by a manager. Of course, being as how he managed for over 50 years (Pittsburgh Pirates 1894-1896 and Philadelphia Athletics 1901-1950) he also holds the MLB record for most career victories by a manager. His career record as a manager was 3,731-3,948.
