It would be very awkward for a race outside of France to be called the Tour de France. And the French would probably have a little something to say about it. The race is essentially a 'tour' of the country.

The majority of the race is held in Mainland France, however each year a few stages are held in neighbouring countries, for example, 2009 they held stages in Spain and Switzerland, 2010 - Holland and Belgium, 2011 - Italy, 2012 - Belgium, 2013 - The French Island of Corsica.

the Tour is a 21-day race with stages of about 200km. It is standard that it travels in most mainland French regions. Every year the Tour has some stages in the French Alps and in the Pyrénées, and the final stage ends on the Champs Elysees avenue in Paris.

The TdF is a long race both in time and distance, far too long to fit in any one city. It'll pass through several cities, and always ends in Paris.
No one city is large enough for it.
