Most people think that it is because of the amount of years he has been playing in the NFL. But if you look back at tapes from his first couple seasons you can see his poise in the pocket. The reason Peyton stays so calm in the pocket is because, first of all, he trusts his offensive line to hold their blocks. Any good quarterback has to trust his line. Look at Mark Sanchez- he has almost no trust in his line, and you can see where it gets him. That's what give Manning the poise in the pocket.
