When King Jamesdied in 1625 he was replaced by his son Charles I. The Puritans became very angry when Charles married Henrietta Maria, a Catholic princess. We also became worried when Catholic lords began to be given positions in Charles court. As a result of these rules large numbers of Puritans moved to North America.
After the Mayflower arrived at the tip of Cape Cod in November, 1620, the Pilgrims explored the coastline in a small boat, to find a suitable place to build their settlement. The place they selected was an abandoned native village called Patuxet, which is now Plymouth, Massachusetts. Having found a site, in December, they sailed the Mayflower across from Cape Cod to Plymouth harbor, where they anchored offshore. (The harbor is very shallow, so they had to anchor the ship more than a mile from shore). They came ashore and began constructing shelter. According to legend, Plymouth Rock was the place where they first set foot on land at Plymouth.
Unfortunately, their voyage to the New World did not result in the kind of success the group wanted, although for a while it seemed they would fulfill their plan for a colony of their own. The Pilgrims, who landed in what is today Plymouth Massachusetts in 1620, were British religious dissenters who believed the Church of England could not be reformed; they wanted to separate from it and start a new life in a theocratic settlement where they could establish a community and also be free to follow their beliefs. By the time their ship landed, they were weakened, hungry, and easily susceptible to disease. A number of the original group died.
But gradually, more Pilgrims arrived, and they built up their new colony. In fact, for a while, there was peace between them and the native people (then referred to as Indians) who already lived there. The Pilgrims were mainly farmers, and by the late 1620s, they were doing well for themselves, raising cattle, and getting fur (helpful in cold winters) from going hunting. The Pilgrims grew their own food and subsisted as they wanted to, while practicing their version of Christianity. Ironically, this group who had sought freedom of religion did not allow any members to question their doctrine or deviate from the official Pilgrim beliefs.
By the late 1630s, good relations between the Pilgrims and the local tribes had broken down, as the natives were upset at how much land the settlers were taking (not just the Pilgrims by this time, but several neighboring colonies as well). The Pequot war occurred in 1637, and other wars and skirmishes followed, including the bloody King Philip's War in 1671-- large numbers of Pilgrim men, and even larger numbers of natives died during that war, and the colony was weakened financially. Ultimately, the Pilgrims' dream for maintaining their own separate colony was dashed when the much larger and more influential Massachusetts Bay Colony was reorganized; the Plymouth Colony was merged into it in 1691.

Somebody else would have. And we wouldn't have Thanksgiving.
One of the main ways it happened was unintentional, when the pilgrims came they brought disease that they had built immunities to but nearly wiped out entire populations of native americans.
Nothing bad happened, the Pilgrims befriended a native American named Squanto who acted as a translator between the Pilgrims and natives. In the Fall of 1621, the Pilgrims shared a feast with the Pokanokets. This meal is reason why Thanksgiving is celebrated.
