As far as history is concerned, at the start of the 2007 season the Arizona Cardinals was the NFL franchise with the most losses in history with 664. Second most was the Detroit Lions with 554 and third most was the Philadelphia Eagles with 533.
Through games played on September 12, 2008 the Phillies franchise has lost 10,095 games which is the most of any MLB team. Sadly it is true. But the Phillies have proved there past wrong and that they are a great team.
If you count them as a sports franchise, the Washington Generals have lost over 13,000 games.
From what I've read, it's California Institute of Technology Basketball. Fairly recently, they snapped their 207-game, 11-year losing streak with an 81-52 win over the Bard College Bears of New York. Apparently their average squad has more high school valedictorians than kids who played high school varsity basketball. Rick Reilly wrote an article in SI about it; it's probably archived online somewhere. The kids from other teams were quoted shaking their hands and saying, "Good job, guys. Now go cure cancer." That pretty much sums it up.
