West for at least 50 miles, then south to the Panama Canal. Head east through the canal, then northeast through the Caribbean Sea to the Atlantic Ocean. (Don't run into any of the reefs, islands, or Florida) Turning north by northwest, it's only about another 1,500 miles to go. Just after passing France, you will want to turn east into the English Channel, if you intend to land at London. Don't forget your sweater and foul-weather gear.

Using this route, I calculate the shortest possible sailing route to be 3550 nm (or 6590 km). If you just want to know the shortest straight line distance then it is 3330 nm (6160 km) from St John's, Antigua to Lands End, UK. (I excluded any of the UK islands since the calculation becomes quite complicated!).
A good route planner is RAC route planner. You chose if you are in a vehicle or walking and set your start and end points. It also includes the number of traffic accidents nationwide and you can add another point on your trip and go from A to B to C instead of just A to B.

If I interpret the question as "What is the fastest sailing route from London TO Wales", the answer is to head south after leaving the estuary, then proceed along the south coast of England to Lands End, thence north to the Bristol Channel. The other routes, via the Caledonian Canal or the north coast of Scotland only become viable options if you are aiming for a circumnavigation of the UK - and only then if you wish to make the passage against prevailing winds.
