What ARE you talking about? There is no such thing as a transmission manifold, . .i would need to know what context this is about to help you.
Auto is a 48 RE manual is a G56
Take out the shifter and pour it directly into the transmission through that opening. Make sure to take the drain plug off to know when it is full. Once fluid starts coming out of the drain hole, you will know it is full. The drain plug is located underneath the transmission, on the driver's side. The bolt has a square shaped head. Good luck.
i don't know if this is exactly an answer but I've noticed mine does that on cold mornings (around 32 degrees)

I don't know what year and which transmission you have but the transmission dipstick itself should have the markings to tell you when it's full when properly read. Anything over the full mark when hot would be considered overfill.

Make sure you don't have a disconnected vacuum hose to the transmission, and that your transmission oil level is full.
