This is of course, a debatable question. The best run I've ever witnessed was Garrison Hearst's 96-yard touchdown against the Jets in overtime on opening day in 1998. First, he made a great cut at the line and broke through the linebackers. Then, he go to the edge and went for the sideline where he laid perhaps the greatest stiff arm in the history of the league to break a tackle. At about the 45-yard line, he cut back, picked up a great block by Terrell Owens of all people, then out ran the defenders down to the 5-yard line. He carried defenders the final five yards into the end zone for the winning touchdown. The run really showcased his all-around ability and strength. At that time, he was one of the greatest backs that ever played. For a couple of years, he was as good as they come.
