No you do not. The only requirement is that you be two years past graduating age. There have been a few players over the years drafted out of semi pro leagues, players hwo never attended college. It is a rare occasion however, but many college players join the NFL before they receive their degree.

The scouts look at the college teams for players and only 1% of them become pro. If not on a college team it would be nearly impossible to get a scout to see you play. The teams don't have an open audition day for tryouts. Stay in school.
